TB-500 is the research-market name for synthetic thymosin beta-4 (Tβ4) and its active fragments. Tβ4 is a naturally abundant human protein whose mechanism has been mapped in detail. That gives the literature an unusual degree of coherence for a research peptide.

Mechanism: actin sequestration

Thymosin beta-4 is a 43-amino-acid peptide and the body's principal G-actin sequestering molecule. It binds monomeric actin and regulates its polymerization into filaments. Actin dynamics drive cell shape, division and movement, placing Tβ4 directly upstream of cell migration. The short central fragment containing the LKKTETQ actin-binding motif reproduces much of this activity; it is the basis of several synthetic versions sold for research.

Outside the ophthalmic programs, Tβ4 research remains preclinical. No injectable TB-500 product is approved for human or veterinary use, and sports anti-doping agencies explicitly prohibit it. Regardless of their consistency, published animal results do not establish human efficacy or safety.
