GHK-Cu Research Overview: The Copper Peptide in Skin, Wound and Gene-Expression Studies
GHK-Cu is a natural human molecule with a five-decade research history. Its copper complex is visibly blue, an unusual property among research peptides.
The molecule
GHK, or glycyl-L-histidyl-L-lysine, is a tripeptide first isolated from human plasma in 1973. It binds copper(II) with high affinity, forming the GHK-Cu complex in which it circulates and acts. One much-cited aging observation is that plasma GHK declines from roughly 200 ng/mL in early adulthood to around 80 ng/mL by age 60. That decline is one reason GHK-Cu became a model compound for skin-aging research.
Main research threads
- Collagen and matrix synthesis: Fibroblast studies report increased collagen, glycosaminoglycan and decorin production. This work forms the basis of GHK-Cu's use in dermatological research.
- Wound models: Rodent and rabbit studies report accelerated closure and improved matrix remodeling, with copper delivery to the wound bed considered part of the mechanism.
- Gene-expression profiling: The most cited modern work comes from screening studies in the Broad Institute Connectivity Map. These studies reported that GHK modulates expression of a substantial fraction of the human genome, including resetting patterns associated with tissue injury toward a regenerative profile. The result is provocative, but these are transcriptomic association studies, not outcome trials.
- Cosmetic formulation research: Topical GHK-Cu is the one commercialized arena, with peer-reviewed facial-study literature reporting improvements in skin density and wrinkle depth versus vehicle.
Limits of the evidence
The cell-culture and animal literature is broad, while controlled human data beyond topical cosmetic studies is thin. No injectable GHK-Cu product holds approval anywhere. The gene-expression findings describe striking transcriptional associations; they do not demonstrate clinical effects.
Practical laboratory notes
The copper complex gives solutions a distinct blue color. This is expected and does not indicate contamination. GHK-Cu is stable in aqueous solution by peptide standards, but normal handling still applies: refrigerate, protect from light, and respect the 30-day window. Avoid co-formulating with strong chelators (EDTA), which strip the copper and change the compound under study.
